Resumo
O DFD-29, uma formulação de minociclina em baixa dose (40 mg), demonstrou eficácia superior no tratamento da rosácea papulopustulosa moderada a grave quando comparado à doxiciclina na mesma dosagem e ao placebo, conforme evidenciado por ensaios clínicos de Fase 3. A análise agregada revelou uma diferença absoluta de 18,0% a 28,3% no sucesso do Investigator’s Global Assessment (IGA) e uma redução adicional de 3,5 a 4,7 lesões inflamatórias em relação à doxiciclina, e a síntese quantitativa dos dados demonstrou superioridade do DFD-29 em relação à doxiciclina, com diferença absoluta de risco de 23,9% (IC 95%: 15,4%–32,4%; p < 0,001), e em relação ao placebo, com diferença de 33,4% (IC 95%: 24,2%–42,7%; p < 0,001), evidenciando benefício clínico consistente da intervenção. Essa vantagem terapêutica é sustentada pela maior lipossolubilidade e potente ação anti-inflamatória da minociclina, enquanto o perfil de segurança mostrou-se favorável, com taxas de eventos adversos comparáveis ao placebo e ausência de efeitos associados a doses antimicrobianas. As concentrações plasmáticas do DFD-29 mantiveram-se consistentemente abaixo do limiar antimicrobiano, reforçando seu perfil subinibitório. Conclui-se que o DFD-29 representa uma opção terapêutica superior e segura, posicionando-se como uma alternativa relevante no manejo dessa condição.
